This paper describes recent information on protein tyrosine kinases and their importance as molecular targets in cancer therapy, as well as the challenges involved in selective binding of drugs while overcoming resistance. Special attention is given to X-ray diffraction studies of enzyme-inhibitor co-crystals, which have demonstrated new binding modes of the second generation of anticancer drugs, tyrosine kinase inhibitors. DOI: 10.5935/1984-6835.2010000
