Aminonucleoside of puromycin is capable of producing the nephrotic syndrome in the rat. 6–mercaptopurine is structurally similar to the purine moiety of the aminonucleoside molecule; because of the similarity and the recent interest in antimetabolite therapy of the nephrotic syndrome, the effect of 6–mercaptopurine on aminonucleoside‐induced proteinuria was assessed in the rat. Concurrent administration of 6–mercaptopurine and aminonucleoside did not cause amelioration of the proteinuria. It is concluded that this observation provides further evidence that immune mechanisms do not play a role in aminonucleoside renal damage, and that 6–mercaptopurine is incapable of competitively inhibiting the action of aminonucleoside.
