Simultaneous determination of the lipoxygenase and hydroperxide lyase specificity in olive fruit pulp

A new radiolabelling\udmethod to determine simultaneously these properties using only\udthin layer chromatography steps is described in the present work.\udThe method involves incubation of an enzyme preparation from\udolive pulp with radiolabelled linoleate, followed by the fractionation\udof the resulting lipid products, previously treated with 2,4-dinitrophenyl\udhydrazine, on thin layer chromatography plates coated with\udpolyethylenglycol400. The results obtained are in agreement with\udprevious studies carried out by other methods.[ES] La regioespecificidad de la lipoxigenasa y la especificidad del\udsustrato hidroperóxido liasa de pulpa de aceituna son parámetros\udimportantes en la justificación de la composición en volátiles del\udaceite de oliva. En este trabajo se describe un nuevo método de\udmarcaje radioactivo para determinar simultáneamente estas propiedades,\udusando sólo etapas de cromatografía en capa fina.
