Identification of Single Genes Controlling Resistance to Stem Canker in Soybean 1

Stem canker [caused by Diaporthe phaseolorum (Cke, & Ell.) Sacc. var. caulivora Athow & Cald.] is a serious disease of soybean [ Glycine max (L.) Merr.] in many areas of the USA. Two major dominant alleles for resistance have been found in ‘Tracy‐M’, each of which conditions complete resistance. However, soybean lines possessing one or the other dominant allele in the homozygous state have not been identified. This has impeded progress in identifying and using other sources of resistance. The objective of this study was to identify lines having one or the other pair of dominant alleles for resistance in Tracy‐M. Crosses were made between Tracy‐M and a very susceptible breeding line, J77‐339. Two selections from that cross were identified as being dominant homozygous for one or the other locus. These were crossed with each other and crossed with J77‐339. Disease reactions in the field and greenhouse confirmed that the two selections possessed homozygous resistance alleles at different loci. The gene symbols Rdc1 and Rdc2 have been assigned.
