Cotyledons of Bromus inertnis Leyss. were fixed and decolorized in Carnoy's solution for 24 hours, immersed in 70% alcohol, and softened in lactic acid at 60 C for 15 min. The whole leaf was then mounted in lactic acid. Mean stomatal lengths of 41.8 and 43.4 µ were obtained for adaxial and abaxial surfaces of cotyledons of tetraploid seedlings compared to 62.3 and 65.1 µ for octoploid seedlings. This simple technique provides a rapid and economical method of screening for polyhaploids in populations of octoploid Bromus inermis .
