The authors take thin pieces of tissue freshly cut with a sharp scalpel and immerse them for 2 or, better, 10 minutes in Schandin's solution (2 parts saturated solution of mercury chloride, 1 part of absolute alcohol, a few drops of vinegar so that the solution contains 4% acetic acid. absolute alcohol and a few drops of vinegar so that the solution contains 4% acetic acid solution), then the preparation is washed in alcohol and distilled water, stained with Mauer's haemalaun and eosin, dried and cleared with absolute alcohol and xylene, after which the sections are embedded in Canadian balsam.
