Synthetic and hydrolytic cholesterol esterase activity of isolated rabbit macrophages was determined in vitro . Homogenates prepared from these cells esterified cholesterol at pH 6·0 and hydrolyzed cholesterol acetate at pH 7·3. It was not possible to demonstrate hydrolysis of cholesteryl oleate by homogenates or to increase the activity by the incorporation of coenzyme A and A.T.P. in the system.
