Electron microscopy using freeze‐fracture replicas revealed denser distribution of PtdIns(3,5)P2 in the vesicular portion (red) than in the tubular portion (green) (upper left panel; HeLa cell). Fluorescence microscopy confirmed distribution of PtdIns(3,5)P2 (red) in the endosome, and further showed its segregation from VPS35, a retromer subunit (green) (upper right panel; COS‐1 cell).
