Molecular Detection and Identification of a 16 S rVI Group Phytoplasma Associated with Tomato Big Bud Disease in X injiang, C hina

In 2010, tomato plants with big bud symptoms were observed in X injiang, C hina. PCR products of approximately 1.2 and 2.8 kb were amplified from infected tomato tissues but not from asymptomatic plants. A comparison of 16 S rDNA sequences showed that the casual tomato big bud ( TBB ) phytoplasma was closely (99%) related to the ‘ C andidatus P hytoplasma trifolii’ (16 S rVI group). The TBB phytoplasma clustered into one branch with the L oofah witches'‐broom phytoplasma according to the 23 S rDNA analysis but with no other member of the 16 S rVI group. The cause of TBB symptoms was identified as ‘ C a . P hytoplasma trifolii' (16 S rVI group) by PCR , virtual RFLP and sequencing analyses. This is the first report of a phytoplasma related to ‘ C a . P hytoplasma trifolii' causing TBB disease in C hina.
