Front cover: Background: Cocaine‐ and amphetamine‐regulated transcript (CART) peptide is a widely distributed neurotransmitter that attenuates cocaine‐induced locomotor activity when injected into the nucleus accumbens (NAc). Main findings: Here we show that cocaine administration enhances the phosphorylated Ca2+/calmodulin‐dependent protein kinaseIIα (pCaMKIIα)‐ D3 dopamine receptor (D3R) interaction in the NAc shell, while intra‐accumbal injection of CART peptide decreases cocaine‐induced locomotor sensitization. Implication: Therefore, we propose that CART peptide exerts its inhibitory effect on cocaine‐induced locomotor sensitization via inhibition of the pCaMKIIα‐D3R interaction, calcium influx and D3R signaling in the NAc shell, which could provide new evidence for the application of CART peptide to treat cocaine‐induced behavioral sensitivity.
