Front cover: Local injection of recombinant adeno associated virus (rAAV) vectors is a powerful tool to manipulate neuronal circuits like spinal sensory circuits. Depending on whether local neurons or neurons providing input to the injection site are to be manipulated different properties of rAAV vectors are required. We have identified promoter/serotype combinations that will either optimize or minimize retrograde transduction while maintaining good local transduction. The photomicrograph shows expression of eGFP in neurons of a dorsal root ganglion (DRG) which have been transduced with a rAAV vector harboring an eGFP expression cassette by intra DRG injection.
