Regenerating tobacco BY‐2 protoplasts are suitable for studying cell polarity and axis formation. In this issue, Zaban et al. (pp. 142‐159) present a model showing the microtubule cytoskeleton conveys positional information between the nucleus and membrane, to control the cell wall formation and the establishment of a new axis.
