Summary The ON / OFF pathways in the retina represent parallel systems with asymmetric properties. Here we discuss the basic anatomy and function of these pathways in health and disease, from the point of view of luminance, contrast and motion perception. We will also highlight how receptive field properties are shaped within each of these pathways and how they cross‐talk within other parallel streams in the retina. Finally, we will address experimental and disease models that dissect the function of each of these pathways.
