SUMMARY A cell‐free system prepared from spleen cells of DBA/2 mice was stimulated by P815 tumour cells as antigen. The response of the cell‐free system to this stimulation was a synthesis of i‐RNA in the cell free system. The i‐RNA was isolated by a combined phenol‐Na‐dodecyl‐sulphate extraction method and purified by gradient centrifugation. This i‐RNA was injected to DBA/2 mice to which P815 tumour cells were implanted at different time intervals before the i‐RNA application. It could be demonstrated that the i‐RNA inhibits the development of tumour. Theoretical and practical implications of the experiments are discussed.
