A tissue culture technique for the study of the mixed leucocyte reaction (MLR) occurring between allogeneic chicken spleen cells was developed. Some features of the one‐way MLR, using mitomycin C‐treated spleen cells as stimulators, and the two‐way MLR were characterized. Cells forming antibody to histocompatibility or other antigens common to lymphoid cells and erythrocytes could not be detected in the MLR. Spleen cells from hypogamma‐globulinaemic bursectomized‐irradiated chickens reacted normally both in the two‐way and one‐way MLR. It is suggested that the MLR occurs between thymus‐dependent lymphoid cells or between such cells and macrophages. Bursa‐dependent lymphoid cells appear to play a non‐essential role, if any, in the MLR.
