Detailed cytological studies of protoplasts isolated from mesophyll tissue of pea leaves were made. The diploid nuclei of mononucleate protoplasts showed normal mitotic division. Binucleate protoplasts showed synchronous nuclear division of two nuclei leading to multinucleate formation. Some of the nuclei of binucleate protoplasts fused prior to nuclear division. The fused nuclei had chromatin connections. Abnormalities in nuclear division were noted in some cases. Protoplasts isolated from older leaves showed a lower frequency of mitotic division than those from younger leaves.
