Ultraviolet (UV) radiation can suppress cutaneous immune responses, thus enhancing susceptibility to skin cancers. The circadian neurohormone melatonin has been shown to have immune-enhancing effects, and to reduce sunburn in mice and humans. Its UV immune protective effects in humans are however unknown. Using the Mantoux model of skin immunity, we tested topical melatonin and its vehicle on healthy volunteers. The lotions were applied to areas on the back daily after each of 3 UV exposures. We found significant and similar levels of immunosuppression at melatonin and vehicle-treated sites. In this model, melatonin did not prevent UV-induced immunosuppression or sunburn.
