Optimal conditions for the assessment of T cell proliferation and cytolytic T lymphocyte (CTL) responses specific for primate lentivirus were established. CTL and T cell proliferative responses were demonstrated as early as two weeks post‐infection. Although the majority of CTL were CD8 + , MHC class I‐restricted, this study demonstrated CD4 + , MHC class II‐restricted CTL. Experiments also demonstrated that CTL were CD 16 negative. Target cell generation could be blocked with CD4 mAb specific for the SIV binding site.
