The Effect of Factor IX and Factor X on Blood Platelets

Human platelet‐rich plasma was filtered through Sepharose 2 B by the method of Tangen & Berman (1972) to obtain platelets free of plasma. Such platelets aggregated normally when 10 ‐5 M ADP or 0.05 U/ml of thrombin were added. When the platelets were exposed to activated factor IX (factor IXa) or activated factor X (factor Xa, EC 3.4.21.6) no aggregation was observed. Even in the presence of cephalin and Ca ++ no aggregation or other changes observed by light transmission occurred. It thus seems that the highly purified coagulation factors IXa and Xa cannot by themselves cause platelet aggregation.
