The influence of several parameters was investigated using modifications of the platelet retention method of Bowie. The results indicated two mechanisms for platelet retention. In von Willebrand's disease platelets were retained in a random manner by increasing the contact time of the platelets. This process was a slow constant retention which in normal individuals was superseded by the appearance of an aggregating principle that rather suddenly markedly increased the platelet retention. This accelerated retention did not occur in the patients with von Willebrand's disease. It appeared faster in columns of long length but was independent of the rate of flow, the amount of blood passed through and the contact time of the platelets. The accelerating principle was probably identical with or closely related to the von Willebrand factor in plasma.
