SYNOPSIS. A small zonal density gradient centrifuge has been designed and constructed for the separation of cell granules from Trypanosoma brucei. Two distinct bands were obtained on a simple sucrose density gradient of the following granules which were found in corresponding fractions of the effluent by electron microscopy: Type I granules (lysosomes) which formed a clear‐cut band toward the periphery of the rotor, and Type II (endoplasmic reticular) granules which formed a more diffuse band and showed great diversity of size, shape and state of aggregation. This aggregation is related to the structure of these granules which are formed in the endoplasmic reticular tubules and are therefore not easily dispersed during rupture of the cell body.
