SUMMARY Methods have been presented for isolation, partial purification, and characterization of DNA from a 7‐chromosome strain of Astrephomene gubernaculilera. Sheath material surrounding the individual cells was removed enzymatically and cells were lysed with hot Na lanryl sulfate. DNA was isolated and purified by a modification of Marmufs procedure. Melting experiments and buoyant‐density values indicated that native DNA with a G‐C content of 61‐62% had been prepared. A pentose phosphorus containing compound was present in stoichiometric amounts.
