Comparative Bioavailability of Phenytoin from Generic Formulations in the United Kingdom

Summary: A multiple cross‐over study was carried out to compare the serum phenytoin concentrations produced in epileptic patients by Epanutin capsules and by four formulations of phenytoin sodium tablets B.P. produced by different manufacturers in the United Kingdom. Although statistically significant differences were found, their magnitude was small and unlikely to cause clinically important alterations in the control of epilepsy. RÉSUMÉ Une étude en cross‐over multiple a été effectuée pour comparer les concentrations sériques en phenytoine obtenues chez des patients épileptiques avec les capsules d'Epanutin à celles obtenues avec quatre formules de comprimés de phenytoine sodique BP fabriquées par differents laboratoires dans le Royaume Uni. Bien qu'il soit apparu des differences statistique‐ment significatives, cellesci étaient faibles et ne pouvaient entrainer d'erreurs cliniquement importantes dans le contrôle de l'épilepsie. RESUMEN Se ha llevado a cabo un estudio múltiple “crossover” para comparar las concentraciones séricas de fenitoína alcanzadas en epilépticos tratados con cap‐sulas de Epanutin y con cuatro formulas de fenitoina B.P. preparadas en laboratories diferentes del Reino Unido. A pesar de que se encontraron diferencias es‐tadisticamente significativas, su magnitud fue pequena y se considero improbable que causaran importantes alteraciones clinicas en el control de la epilepsia. ZUSAMMENFASSUNG Es wird von einer mehrfachen cross‐over Studie be‐richtet, bei der Phenytoinkonzentrationen im Serum von Epileptikern bestimmt werden nach der Einnahme von Epanutinkapseln und Phenytoinnatriumtabletten B.P., die von verschiedenen Firmen im United Kingdom hergestellt werden. Obwohl statistisch signifi‐kante Unterschiede gefunden wurden, ist ihr Ausmaß gering, und es erscheint unwahrscheinlich, daß sie klinisch wichtige Ändefung bei der Kontrolle von Epilepsien bewirken können.
