The Delayed Effect of Sodium Valproate on the Photoconvulsive Response in Man

Summary: The acute effect of a single oral dose of sodium valproate on the photoconvulsive response has been studied. In 7 of 9 patients naive to this drug photosensitivity was abolished or reduced. A similar effect was seen in only 2 of 7 already receiving chronic sodium valproate therapy. The effect appeared 1 to 5 hr (mean, 3 hr) after attainment of peak sodium valproate concentration and lasted up to 5 days. RÉSUMÉ Ľeffet d'une dose orale unique de valproate de sodium sur la résponse photo‐convulsive àétéétudié. Sur 9 patients qui n'utilsaient pas preéalablement ce médicament, la photosensibilité a été supprimée ou réduite chez 7 d'entre eux. Le méme effet n'a été observe que 2 fois chez 7 sujets déjà soumis à l'effet du médicament de façon chronique. L'effet apparait 1 à 5 heures (en moyenne 3) après que le taux sérique du valproate de sodium ait atteint son maximum et il persiste jusqu'a 5 jours. RESUMEN Se ha estudiado el efecto agudo de una dosis unica oral de valproato sódico sobre la respuesta fotoconvulsiva. En 7/9 pacientes que no habfan tornado esta medicación, se abolió o se redujo la fotosensibilidad. Se observó un efecto seme jante en solo 2/7 que ya recibfan un tratamiento crónico con valproato sódico. El efecto apareció entre 1 a 5 horas (promedio de 3) después de alcanzarse el máximo de concentración de valproato sódico y duró hasta 5 días. ZUSAMMENFASSUNG Die akute Wirkung einer einzelnen oralen Dosis Natrium‐Valproat auf die fotokonvulsive Reaktion wurde untersucht. Bei 7 von 9 Patienten, die erstmalig dieses Medikament bekamen, wurde die Fotosen‐sibilität reduziert oder zum Verschwinden gebracht. Eine vergleichbare Wirkung wurde nur bei 2 von 7 Patienten beobachtet, die bereits Natrium‐Valproat chronisch erhielten. Die Wirkung trat 1 bis 5 Stunden im Schnitt 3 Stunden nach Erreichen der maximalen Natrium‐Valproat Konzentratipn auf und dauerte bis zu 5 Tagen.
