The Effects of GABA and Strychnine on Neocortical Structures involved in the Onset Mechanism of Epileptic Activity

SUMMARY The changes in convulsive reactivity to direct neocortical stimulation, produced by GABA and strychnine, applied topically, either drug alone, or both combined, were studied in unanaesthetized cats. GABA results in no change of convulsive reactivity. Strychnine applied to a GABA‐soaked neocortex enhances the convulsive reactivity to direct stimulation in the same manner as strychnine alone. In experiments dealing with GABA and strychnine action on single responses elicited by liminal and supraliminal direct cortical stimulations, on responses evoked by peripheral stimuli or by single shocks in the lateralis posterior nucleus of the thalamus, the mechanisms of action of these drugs on the neocortical functions were investigated. It is concluded that GABA, applied topically, acts as a depressant of neocortical superficial structures, permitting thereby a hyperexcitability of the deeply situated structures and that strychnine acts especially on the internuncial neurons. Le GABA n'entraine aucune modification de la reAactiviteA convulsive. La strychnine appliqueAe sur le cortex deAjaG imbibeA de GABA augmente la reAactiviteA convulsive de lameCme manieGre que la strychnine seule. L'auteur eAtudie ensuite le meAcanisme d'action du GABA et de la strychnine sur le fonctionnement neAocortical en appreAciant l'effet de ces produits sur les reAponses isoleAes provoqueAes par les stimulations corticales directes liminaires et supraliminaires et sur les reAponses eAvoqueAes par les stimuli peAripheAriques ou des chocs isoleAs appliqueAs dans le noyau lateralis posterior du thalamus. La conclusion est que le GABA appliqueA localement posseGde une action deApressive sur les structures neAocorticales superficielles, permettant ainsi une hyperexcitabiliteA des structures profondes, tandis que la strychnine agit speAcialement sur les neurones internuntiaux.
