Kynurenine formamidase (aryl‐formylamine amidohydrolase, EC 3.5.1.9) was found to be present in rat brain and was partially purified and characterized. The partially purified enzyme catalysed the hydrolysis of 5‐hydroxyformyl‐ dl ‐kynurenine to 5‐hydroxy‐ dl ‐kynurenine and that of formyl‐ l ‐kynurenine to l ‐kynurenine at similar rates. The apparent K m values of the enzyme for 5‐hydroxyformyl‐ dl ‐kynurenine and formyl‐ l ‐kynurenine were 4.0 ± 10 −4 and 1.8 ± 10 −4 m , respectively. The enzyme was active over a wide pH range (5.5–8.5). The activity was inhibited by low concentrations of Ag + and Hg 2+ . The physiological significance of the enzyme is discussed.
