SUMMARY The preparation of genomic DNA from a recently isolated group of cyanophage which infect species of Nostoc and Anabaena (cyanobacteria) has been investigated. Factors which affect the yield of DNA, including culture density on infection, divalent cation requirements, NaCl tolerance and desorption from cell debris, have been assessed. These factors have been incorporated into optimized procedures for the isolation of genomic DNA from five cyanophage. The DNA isolated from the rive cyanophage species was identified using restriction endonuclease fragment patterns.
