SUMMARY The possibility of preparing plant mitochondria using Sephadex or Bio‐gel columns was investigated. It is concluded that gel filtration is a feasible alternative to centrifugation with many plant materials. It has the advantage of providing preparations of higher O 2 uptake rate per unit protein with similar or better ADP/O or respiratory control ratios than centrifugation. It is often also more rapid for routine preparation. Its disadvantage in particular is the dilution of the final suspension.
