Summary— The action of diethylstilboestrol on the androgen response mechanism of the human prostate has been examined. A simple in vitro test system that assesses effects on hormone uptake, receptor binding and translocation to the nucleus has been employed. The test has also been modified to study the effect of diethylstilboestrol on the activity of 5α‐reductase, the enzyme responsible for the conversion of testosterone to 5α‐dihydrotestosterone. These results are discussed in relation to the dosage of diethylstilboestrol for treatment of carcinoma of the prostate.
