Summary: Chlorpromazine and other tranquillizers cause a precipitation of the protein and glycoprotein components of bile in vitro . This precipitation depends on an interaction between the carboxyl groups on the protein and glycoprotein components of bile and the positively charged amine groups on the drug molecules in solution. It is suggested that administration of the tricyclic anti‐depressants and tranquillizers may result in a sufficient concentration of these drugs or their metabolites in bile to cause some precipitation of the bile components within the bile canaliculi. Such a precipitation could play a role in the development of the cholestatic jaundice which occurs as a side effect in patients being treated with these drugs.
