Early Occurrence of Alzheimer's Neurofibrillary Tangles (NFT) in Developmental Brain Damage and Chronic Brain Diseases with Early Childhood Onset

We observed early occurrence of neurofibrillary tangles (NFT) in brains with various types of developmental brain damage or chronic neurologic disorders starting at infancy or early childhood. The diseases included Fukuyama congenital muscular dystrophy (FCMD), porencephaly, postencephalitic epilepsy, hydrocephalus, Cockayne's syndrome and Hallervorden‐Spatz disease. The earliest NFT were observed in a 12‐year‐old girl with congenital toxoplasmosis, but most of the patients were over 20 years of age. NFT frequently occurred in the cerebral cortex, nucleus basalis of Meynert and brain stem nuclei, but were not common in the hippocampus. Senile plaques were absent in all cases. We discuss the significance and hypothesis of NFT occurrence in such patients.
