Four DNA extraction protocols were investigated to identify a protocol that optimizes extraction of DNA from onion. Allium cepa , for ampli| fication of DNA by polymerase chain reaction. The protocol selected was a simple procedure which used fresh leaf tissue, resulted in high UNA yields and low DNA degradation, low RNA contamination. was the least costly and required only 45 min. When RNAase T1 rather than RNAase A was used, more RNA was eliminated.
