Culture of Trichophyton verrucosum in a liquid medium

Summary The possibility of application of Sabouraud̂s liquid medium for isolation and multiplication of Trichophyton verrucosum was investigated. It was found that the use of the liquid medium for isolation of the fungus from the infected material produced the growth of T. verrucosum within 9–11 days; the medium did not get dry during the cultivation of the organism, was cheap, and enabled isolation of the fungus even in cases of contamination of the culture with fungi of the Aspergillus sp. The liquid medium was also useful to multiply T. verrucosum. All 27 examined strains of T. verrucosum grew in Sabouraud̂s medium within 9 days. The above technique produced a large mass of the fungus; this fact is of great importance for antigen and vaccine preparation, to make monovalent trichophytin etc. T. verrucosum cultivated in Sabouraud̂s liquid medium did not lose its virulence for guinea‐pigs. Zusammenfassung Die Möglichkeit der Verwendung von fliissigem Sabouraud‐Nährboden zur Isolierung und Vermehrung von Trichophyton verrucosum wurde untersucht. Es wurde festgestellt, daβ der Gebrauch dieses Nährbodens zur Züchtung des Pilzes aus infektiösem Material in 9 bis 11 Tagen führte. Der Nährboden—billig in seiner Anwendung—trodmete während der Züchtung des Dermatophyten nicht aus. Die Anzüchtung des Pilzes war sogar bei Verunreinigung der Kulturen durch Aspergillus‐Arten möglih. Dieser flüssige Nährboden eignet sich gleichfalls zur Vermehrung von T. verrucosum. Alle 27 untersuchten T. verrucosum‐Stänime verloren ihre Virulenz für Meerschweinchen nicht.
