A procedure for efficient isolation and cell wall regeneration of protoplasts from Botrytis cinerea is described. Protoplasts were obtained from mycelia using a lytic enzyme mixture containing β‐Glucuronidase, Cellulase R10 and Driselase with mannitol for osmotic support. The digestion of cell walls was checked by fluorescence microscopy. Protoplasts were purified from cell debris and lytic enzymes. Regeneration and reversion were performed by incubation on agar plates.
