Method for the demonstration of resting spores of the clubroot causing fungus Plasmodiophora brassicae in horticulture soil samples The investigation based on the biochemical determination of the polymer chitin for a specific demonstration of the clubroot causing fungus Plasmodiophora brassicae. For separation of resting spores from soil material a special apparatus has been constructed which allows the continuous isolation and concentration of this fungal developmental stage. The specificity of the test is improved by a differential density gradient centrifugation. N‐acetyl glucosamine as the measurable reaction product was formed by chitinase. Only two days are necessary for the whole test procedure. Its sensitivity is 2.5 × 10 7 resting spores/litre soil material. The method allows a first screening of soil samples and increases the efficiency of common biological tests.
