The method is based on the capacity of many fungi to invade and survive in wood for a long time. Small standard pieces of inoculated wood are piled up in test tubes stored in cold conditions. Reisolation of the fungus is easy by picking out only one piece of wood without disturbating the whole arrangement. By this process a large number of fungi, not only basidiomycetes, can be stored over many years without any subculturing.
