Rat liver chromatin was sedimented in a sucrose density gradient under alkaline conditions. It is shown that in the pH interval 12.2—12.8 four histone species are dissociated stepwise in the sequence: F2b, F1, F2al and F3. Histone F2a2 remains bound to DNA even at pH 13. About 70% of the non‐histonc proteins dissociate from the chromatin at pH values lower than 12.2 and 30% remain bound to DNA in the range of pH studied.
