Incorporation of 5‐Iodocytidine into Yeast tRNA Phe with tRNA Nucleotidyl Transferase in vitro

5‐Iodocytidine (io 5 C) is incorporated into tRNA Phe ‐A 73 as well as into tRNA Phe ‐A 73 ‐C 74 using tRNA nucleotidyl transferase. The tRNA Phe ‐A 73 ‐C 74 ‐io 5 C 75 ‐A 76 and tRNA Phe ‐A 73 ‐io 5 C 74 ‐io 5 C 75 ‐A 76 , respectively, thus obtained can be aminoacylated by the corresponding synthetase exhibiting the same K m but a slightly lowered V compared to tRNA Phe ‐A 73 ‐C 74 ‐C 75 ‐A 76 . An analytical method is presented which enables quantitative determination of the 3′ terminal nucleosides.
