The effect of chemical inhibitors in two human in vitro systems for detecting antibody‐dependent (ADLC) and direct lymphocyte‐mediated cytotoxicity (DLC) was studied. Both reactions were inhibited by cytochalasin B and by depletion of divalent cations. The immunosuppressant methylprednisolone inhibited both reactions. No inhibition was found with 6‐mercaptopurine. DLC was inhibited by puromycin and actinomycin D whereas ADLC was not. No inhibition was found with mitomycin C.
