By amino acid analyses and radioactive titration of cleaved S–S bonds on reduction, it was shown that the hinge region of IgG3 and its F(ab′) 2 fragment seem to contain 15 S–S bridges. About three of the bridges are not reduced by 0.01 M dithiothreitol in aqueous buffer, and, in addition, this treatment cleaved the F(ab′) 2 fragment into two Fab′ subunits. The observation, therefore, may support the idea that the γ 3 ‐chain contains an extra intrachain disulphide loop.
