Electron phase‐contrast imaging of melamine‐embedded, unstained horse‐spleen ferritin

SUMMARY A pellet of aldehyde‐fixed horse‐spleen ferritin was embedded in a water‐soluble melamine resin for ultrathin sectioning. Overfocused electron micrographs of individual molecules show the four‐lobed iron‐core and the peripheral protein subunits in great detail. They confirm earlier X‐ray diffraction data of hydrated horse‐spleen apoferritin (Hoare et al., Nature , 255, 653, 1975), which postulate a molecule with twenty‐four protein subunits occupying octahedral lattice sites around the iron‐core.
