Action of gibberellic acid in a dwarf maize mutant: an X‐ray microanalysis study

SUMMARY A method is described to prepare plant tissue for X‐ray microanalysis. Model experiments with radioactively labelled ions show that freeze substitution can be used for the retention of soluble ions under controlled conditions. For calculations of absolute values a combination of the ratio method and the continuum method is used with dentine as the only standard. An X‐ray microanalysis study of the action of plant hormones was performed with a dwarf maize mutant, sensitive to minimal amounts of the plant hormone gibberellic acid (GA 3 ). The dwarf corn mutant shows completely different concentrations of K + , Ca ++ and Cl in chloroplasts, cytoplasm and vacuoles in comparison with the normal plant. GA 3 produces changes of the ion concentrations similar to those of the normal plant. Short‐term experiments (1‐4h action of the hormone) also result in changes in the ion concentrations, specifically due to the hormone. The structurally similar by physiologically inactive GA 3 methylester causes a response comparable to that of water.
