SUMMARY A technique for preparing human skin for high‐resolution scanning electron microscopy using phosphate‐buffered crude bacterial α‐amylase is described. Effects of the buffer solution alone are studied as well as the proteolytic activity of the enzyme solution used. The biochemical implications of these observations are discussed with regard to the in situ structure of the tissue.
