Summary An investigation was made of cultural conditions affecting the blackening of colonies due to sulphite reduction by Clostridium welchii and several other clostridia in the sulphite‐polymyxin‐sulphadiazine agar of Angelotti et al. (1962). Reducing conditions in the medium were more critical for blackening than for growth, and a simple, modified isolation method for Cl. welchii is described which provides optimal conditions for both processes.
