S ummary A functional Krebs tricarboxylic acid cycle was demonstrated to exist in rabbit reticulocytes by the following methods:1 Measurement of the rate of oxygen consumption of the cells, using acetate and succinate as substrates. 2 Demonstration of the Crabtree effect exerted by glucose on cellular respiration. 3 Following the metabolic pathway of [2‐14 C]acetate, [3‐14 C]pyruvate and [6‐14 C]glucose and measuring the formation of labelled CO2 by the cells. The effects of some inhibitors of mitochondrial activity on this pathway were also studied. 4 Demonstration of the conversion of labelled acetate into various intermediates of the Krebs cycle by chromatography and autoradiography.
