SUMMARY Platelet production is stimulated by removal or destruction of platelets and is inhibited by hypertransfusion with viable platelets. It is not influenced by factors regulating red cell production, even in the absence of the spleen. Nephrectomy does not impair the response to platelet depletion. It is concluded that the regulation of platelet production is by a system entirely independent of that governing erythropoiesis.
