Freeze‐fracture replication of X‐granules in cells of cutaneous lesions of histiocytosis‐X (Letterer‐Siwe disease)

summary Fresh, not chemically fixed, glycerol‐cryoprotected material from skin lesions of histiocytosis‐X (Letterer‐Siwe disease) was examined by the freeze‐fracture technique. Fracture appearances of the cytoplasmic X‐granules characteristic of cells of histiocytosis‐X were established, and the accuracy of previous three‐dimensional reconstructions of variations, in overall shape of granules based upon stained thin sections was confirmed. These variations, therefore, are not artifacts of conventional tissue processing. Little or nothing of the internal structure of X‐granules is seen in replicas, and the technique provides no new information concerning their origin or mode of formation.
