A sliced tumor of dermatofibrosarcoma protuberans was labelled with 3 H‐glucosamine in Ham's F12 medium with 10% fetal calf serum at 37°C in 5% CO 2 with 95% air for 24 hours. Electrophoresis of extracted glycosaminoglycan was undertaken on cellulose acetate strips. The radioactive content of the separated glycosaminoglycan in the tumor was counted and compared with the glycosaminoglycan of normal skin. A larger amount of hyaluronic acid and chondroitin sulfate was synthesized in dermotofibrosarcoma protuberans than in normal skin. The qualitative and quantitative changes in glycosaminoglycan in the stroma suggest that it is unlikely to metastasize to the regional lymph nodes and internal organs.
