Using structure‐based combinatorial engineering approaches, ubiquitin variants (UbVs) are selected through phage display as modulators of various protein–protein interactions in the ubiquitin signaling pathway. Here, we compile the recent progress using UbVs as inhibitors, activators, or biochemical probes for different families of human E3 ligases. We propose the future application of UbVs in inducing proximity to E3 ligases for targeted protein degradation and developing therapeutic agents directly targeting E3 ligases.
