Prosaptide™D5, a retro‐inverso 11‐mer peptidomimetic, rescued dopaminergic neurons in a model of Parkinson's disease

In this report we present a novel strategy for the rescue of dopaminergic neurons in a Parkinson's disease (PD) model by using a bioactive protease resistant RETRO‐INVERSO trophic peptidomimetic derived from prosaposin, Prosaptide™D5. When injected subcutaneously in mice with MPTP toxicity, Prosaptide™D5 treatment rescued DA neurons in the substantia nigra; the rescue was complete 15 days after treatment. Treatment with Prosaptide™D5 also rescued neurons from cell death IN VITRO by increasing neurite sprouting and by normalizing dopamine (DA) uptake. Prosaptide™D5 is a unique peptidomimetic trophic factor that has been shown to be effective on a PD model when given systemically.
